The eastern English Channel, which connects the Atlantic Ocean to the North Sea through the Dover Strait, is an area where numerous and often conflicting human activities take place. A cross-border multidisciplinary project called CHARM was initiated to provide knowledge and tools for planners and decision-makers to durably manage the shared marine living resources. One such tool was an atlas of fish spatial distributions and modelled habitats, which was used here to investigate ontogenic and seasonal shifts in fish spatial distribution and habitat through a case-study, the dab Limanda limanda. Survey data for several life-history stages (eggs, larvae, coastal nurseries, < and > 1 year old) and seasons were used to map spatial patterns (using...

Comparison of traditional microscopy and digitized image analysis to identify and delineate pelagic fish egg spatial distribution ArchiMer
Lelievre, Stephanie; Antajan, Elvire; Vaz, Sandrine.
One of the problems concerning studies of fish egg distribution is the weak spatial and temporal resolution due to the workload that examination of a large number of samples would demand. Recently, the development of a new laboratory imaging system, the ZooScan, capable of obtaining relatively good resolution images enables automated zooplankton identification using supervised learning algorithms. This new approach was applied to formalin-fixed fish egg samples collected during French winter IBTS (International Bottom Trawl Surveys) in the Eastern English Channel and the Southern North Sea. Fish egg spatial distributions of seven species based on the microscope and ZooScan identifications were compared. Abundance and distribution maps of winter-spawning...

Delineating recurrent fish spawning habitats in the North Sea ArchiMer
Lelievre, Stephanie; Vaz, Sandrine; Martin, C. S.; Loots, Christophe.
The functional value of spawning habitats makes them critically important for the completion of fish life cycles and spawning grounds are now considered to be “essential habitats”. Inter-annual fluctuations in spawning ground distributions of dab (Limanda Limanda), plaice (Pleuronectes platessa), cod (Gadus morhua) and whiting (Merlangius merlangus) were investigated in the southern North Sea and eastern English Channel, from 2006 to 2009. The preferential spawning habitats of these species were modelled using generalised linear models, with egg distribution being used as proxy of spawners’ location. Egg spatial and temporal distributions were explored based on six environmental variables: sea surface temperature and salinity, chlorophyll a concentration,...

Identification and characterisation of winter spawning ground in the English Channel and Southern North Sea ArchiMer
Lelievre, Stephanie.
A better knowledge and monitoring of principal commercial fish spawning grounds have become necessary in the North Sea. The efficiency of CUFES was proved by sampling pelagic fish eggs in winter in Eastern Channel and Southern North Sea. Fish egg taxonomic identification based on visual criteria cannot always be carried out effectively. In particular, cod (Gadus morhua), and whiting (Merlangius merlangus) or flounder (Platichthys flesus) and dab (Limanda limanda) have the same range of egg diameter and similar morphologies. Alternative identification methods using molecular techniques were developed to improve the accuracy of egg taxonomic identification. First, PCR-RFLP method, then, in order to accelerate egg identification, the use of a new laboratory...

Integrating molecular identification of pelagic eggs with geostatistical mapping to improve the delineation of North Sea fish spawning grounds ArchiMer
Lelievre, Stephanie; Jerome, Marc; Maes, Gregory E.; Vaz, Sandrine; Calaivany, Sachidhanandam; Verrez-bagnis, Veronique.
Maps of the spawning grounds of commercially important fishes are necessary when assessing the level of connectivity between life stages of fishes and for identifying ecologically valuable marine areas. A first step toward mapping the spawning grounds is a reliable and rapid species identification of pelagic fish eggs to assess the spatio-temporal distribution of spawning aggregations. As many species have similar egg sizes and morphology, the molecular validation of visually identified eggs is often essential for the use of such data in fisheries management. In the present study, we developed a rapid 16S rRNA PCR-restriction fragment length polymorphism (RFLP) assay to distinguish between formalin-fixed fish eggs of dab Limanda limanda, flounder...

ISADO - Identification des frayères du Détroit du Pas-De-Calais et des zones marines adjacentes ArchiMer
Lelievre, Stephanie; Vaz, Sandrine; Martin, Cs.
Spawning is an essential element of the life cycle of marine fish. Fish eggs present a high natural mortality, which makes them a particularly vulnerable stage of fish development. With more and more fish species becoming over-exploited, commercial fisheries are increasingly sensitive to fluctuations in the recruitment of young individuals to the spawningstock. Egg data were collected in the southern part of the North Sea and Dover Strait using a Continuous Underway Fish Egg Sampler(CUFES). With co-funding from the Interreg IIIa Programme, the ISADO (Identification of the Spawning Areas in the Dover Strait and adjacent marine areas) project processed800+ egg samples, with the development of a taxonomic identification key. The spawning areas of seven fish...

Linking spawning ground extent to environmental factors - patterns and dispersal during the egg phase of four North Sea fishes. ArchiMer
Hoffle, Hannes; Van Damme, Cindy J. G.; Fox, Clive; Lelievre, Stephanie; Loots, Christophe; Nash, Richard D. M.; Vaz, Sandrine; Wright, Peter J.; Munk, Peter.
Previous studies have shown that four commercially important demersal species, namely cod (Gadus morhua), haddock (Melanogrammus aeglefinus), whiting (Merlangius merlangus) and plaice (Pleuronectes platessa) spawn in distinct areas across the North Sea. Based on two comprehensive ichthyoplankton surveys in 2004 and 2009, the present study uses Generalized Additive Mixed Models (GAMMs) to delimit these spawning grounds using the distribution of recently spawned eggs, investigates their relationship to specific environmental conditions and examines egg dispersal during their development. Results indicate that presence/absence of early stage eggs is more related to temporal and topographic variables, while egg densities are closely linked with hydrography....

PCR-RFLP analyses of formalin-fixed fish eggs for the mapping of spawning areas in the Eastern Channel and Southern North Sea ArchiMer
Lelievre, Stephanie; Verrez-bagnis, Veronique; Jerome, Marc; Vaz, Sandrine.
A better knowledge on spawning grounds of principal commercial fishes from North Sea is necessary for their monitoring. Fish egg taxonomic identification based on morphological characters cannot always be carried out effectively. This is particularly the case for a number of Gadidae fish species eggs (i.e. cod, haddock and whiting, and three rockling species) which have the same range of size and for which molecular-based techniques can help improving the accuracy of egg taxonomic identification, and hence the reliability of studies on spawning areas. In this study, formalin fixation, egg conservation and DNA extraction were optimized in order to develop a specific PCR-RFLP method. Based on the sequence of the cytochrome b gene of reference specimens, a...

The effectiveness of the continuous underway fish egg sampler compared with standard vertical plankton hauls for sampling the eggs of demersal species in the southern North Sea ArchiMer
Lelievre, Stephanie; Vaz, Sandrine; Fox, Christopher.
To date, the continuous underway fish egg sampler (CUFES) has been used primarily to map the spawning areas of pelagic fish in upwelling regions. To assess its utility in mapping the spawning grounds of demersal species, species composition and densities of eggs collected by CUFES were compared with results obtained from ring-net samples (VET: vertical egg trawl) from the southern North Sea over three annual cruises conducted in January/February. CUFES collected slightly fewer eggs per unit volume of water filtered when compared with vertically integrated samples but maps of egg distributions from both devices showed broadly similar patterns. Geostatistical analyses revealed that small-scale variability was greater with CUFES than with VET. CUFES may thus...
